Some unpublished results are also reported and discussed. 8 ABSTRACT Characteristic features in asthma and chronic obstructive pulmonary disease (COPD) are airfl ow limitation and bronchial infl ammation. During airway infl ammation, increased numbers of activated cells (e.g. eosinophils and neutrophils) associated with infl ammation are found in bronchial mucosa, in bronchoalveolar lavage (BAL) fl uid, in sputum, and, in severe cases, in blood. Both eosinophils and neutrophils contain various mediators in their intracellular granules. When such cells are activated, various types of harmful mediator are released into the immediate environment. Until recently, investigation of airway infl ammation has mainly relied on monitoring of lung function. However, numerous bronchial biopsy and BAL fl uid studies have revealed that airway infl ammation can occur in the absence of bronchial hyper-reactivity, if respiratory disease is mild or clinically well controlled. Cellular and biochemical accompaniments of airway infl ammation have been widely studied by investigation of BAL fl uid and bronchial …

The effect of dithiothreitol on VLA-4 detection in peripheral blood and induced sputum cells.

BACKGROUND Induced sputum has been shown to be a reliable technique for investigating airway inflammation non-invasively. Flow cytometry could provide useful information in this area. However, the viscosity of the sample entails the use of a mucolytic agent. Dithiothreitol (DDT) is the most frequently used agent although it could affect detection of different inflammatory markers. METHODS To ...

Eosinophil Activation Markers in Induced Sputum in Asthmatics

OBJECTIVES Eosinophils play an important role in asthmatic airway inflammation collaborately with other inflammatory cells. The present study was aimed to determine whether the eosinophil activation markers in induced sputum reflect the clinical status in asthmatics. METHODS The clinical severity and FEV1 were measured. Hypertonic saline induced sputum was obtained from 25 asthmatics and ten ...

Long-term repeatability of induced sputum cells and inflammatory markers in stable, moderately severe COPD.

STUDY OBJECTIVES Neutrophilic inflammation is a major feature of COPD. Induced sputum is increasingly used to monitor inflammatory airway diseases. Although short-term repeatability of selected sputum markers has been extensively studied in several populations, data on the long-term repeatability of induced sputum markers in stable COPD are scant. DESIGN Sputum supernatant of 12 patients with...

Expression of the Apoptosis Inhibitor Bcl-2 in Sputum Eosinophils in Patients with Asthma

Background and Objectives: Apoptosis of eosinophils is of significant value in assessing the airway inflammation in patients with asthma. Our purpose was to investigate the degree of expression of the Bcl-2 protein in sputum eosinophils during acute asthma exacerbation and its relationship with exacerbation severity.    Materials and Methods: The study was carried out in Mostafa Khomeini Ho...

Noninvasive assessment of airway inflammation in children: induced sputum, exhaled nitric oxide, and breath condensate.

Noninvasive markers of airway inflammation are needed for use in research and clinical practice in childhood asthma. Induced sputum and exhaled nitric oxide are well established as direct markers of inflammation for use in asthma research. Sputum can be induced from children of >6 yrs using inhalation of hypertonic saline, and, if appropriate, can be combined with an assessment of airway respon...
